چکیده

We have examined the impact of partial injury to central noradrenergic terminals on whole tissue norepinephrine (NE) content in hippocampus and on the concentration of NE and 3,4-dihydroxyphenylacetic acid in extracellular fluid of that structure (using microdialysis perfusion). Partial unilateral depletions of hippocampal tissue NE content were produced by administration of 6-hydroxydopamine (2-10 micrograms) into the dorsal noradrenergic bundle, and 2 weeks later microdialysis probes were placed in hippocampus ipsilateral to the lesion. The resting concentration of NE in hippocampal dialysates was unaffected by the lesion unless the reduction of tissue NE content exceeded 50%. In contrast, the basal concentration of 3,4-dihydroxyphenylacetic acid in hippocampal dialysates declined in proportion to tissue NE content. Tail shock or local perfusion with excess K+ increased NE in dialysates from sham-lesioned animals and produced equivalent changes in NE in dialysates from animals with moderate (less than or equal to 50%) depletions of tissue NE content. No significant increases of NE in dialysates were observed in response to these stimuli in animals with depletions of tissue NE content greater than 50%. To the degree that transmitter level in dialysates is representative of extracellular transmitter concentration, the results suggest that compensatory processes exist by which a normal extracellular concentration of transmitter can be maintained under both basal and stimulated conditions despite the loss of up to one half of a neuronal population. The effects of tramadol on norepinephrine and MHPG releasing in locus coeruleus in formalin test in rats: a brain stereotaxic study

Objective(s):The relationship between tramadol, as an antinociceptive drug, and locus coeruleus (LC), the main noradrenergic nucleus of the brain that affects regulation and modulation of pain through descending noradrenergic pathways was investigated. Materials and Methods: Male Sprague-Dawley rats were divided into four groups of 10 rats. The rats were fixed in stereotaxic instrument and then...

Effect of Norepinephrine depletion on induction of experience dependent plasticity in male rat barrel cortex

Introduction: Barrel cortex of rats is a part of somatosensory cortex, which receives information from facial whiskers. Vibrisectomy by sensory deprivation leads to some changes in the barrel cortex, which have been known as experience dependent plasticity. On the other hand, Norepinephrine (NE) and locus coeruleus, which is the main source of NE, influenced response properties of cortical bar...

The relationship between discharge rates of locus coeruleus noradrenergic neurons and rates of norepinephrine release was examined in the anesthetized rat. Neuronal discharge rates of locus coeruleus neurons were altered and quantified using a combined recording-infusion probe. Peri-locus coeruleus infusions of either the cholinergic agonist, bethanechol, or the a2-agonist, clonidine, were used...

Differential sensitivity of intranuclear and systemic oxytocin release to central noradrenergic receptor stimulation during mid- and late gestation in rats.

A number of changes occur in the oxytocin (OT) system during gestation, such as increases in hypothalamic OT mRNA, increased neural lobe and systemic OT, and morphological and electrophysiological changes in OT-containing magnocellular neurons, suggestive of altered neuronal sensitivity, which may be mediated by ovarian steroids. Because central norepinephrine (NE) and histamine (HA) are potent...

The effect of intracerebroventricular injection of CGRP on pain behavioral responses and monoamines concentrations in the periaqueductal gray area in rat

Objective(s): Calcitonin gene related peptide (CGRP) receptors are widely distributed in the central nervous system. The aim of this study was to investigate the effects of intracerebroventricular (ICV) injection of CGRP on pain behavioral responses and on levels of monoamines in the periaqueductal gray area (PAG) during the formalin test in rats.Materials and Methods: Twenty-four male rats wer...
